Yes,
I am running a 302. My fuel supply line goes up the drivers side of the car and the return line comes back on the passenger side. Both along the main rails. I have the B&B tank in back (an aftermarket polyurethane fuel cell). It has a return line entrance to it. If your tank does not have one, I would run it to the suction side of the pump. If your pump is in-tank, then you will need to punch a hole in the tank somehow. A small bit of welding would be required here.
