Shifter locations different ?
Tranny crossmember different location ?
I'm a retired dirt sprint car racer, so I need power, 600-700 HP/TQ. My driving style is mixed fun.
What's your thoughts on a tranny ?

The T-56 Magnum is roughly 4" longer than the TKO, though the input shaft is about 1.5" shorter so you gain some of that back. The tranny has multiple shifter options / locations. http://www.tremec.com/anexos/File/TR...erformance.pdf
Quicktime has a couple of bellhousing options to mate the T-56 Magnum to a 385. McLeod apparently also has an option - article here on a T-56 into a '71 Mustang: Mustang C6 to T56 Transmission Swap - FordMuscle

1st gear is a parade gear, and 2nd gear starts are easy with a big block.
Mild build 460 - 400 HP / 500 torque.
$3,500 would be a current ballpark price - the transmission was in the car when I bought it in 2014, and the car was built in 1991. Roughly 25,000 miles on it.

Knowing what engine you're hoping to install Kev I'd steer away from any TR3550s and T5s, just no where near enough of a high torque rating. That leaves the TKO600, T56 Magnum or a B/B top loader. I'd have mentioned the TKO500 but you don't need that deep of a first gear for a big block, especially at the cost of 100 lb ft less torque rating. I've a TKO600 in mine and couldn't be happier with it.

TKO, TKO-600RR, Top Loader
I had a 428 with a TKO-3550 and 3.50 locker in a 67 mustang about 15 years ago. It was a bit tight - hard to shift fast. 5th gear was a little too high.
My Cobra came with a top loader and 3.54 Jag rear end. The gear ratios were off. 1st gear wasn't low enough. 4th was too low. Probably has the 2.32 first. I never checked. Highway cruising was painful.
I switched to a TKO600RR. I was worried about the tightness from my earlier TKO, so I got the one modified by Brent Lykins. I am very impressed with how well this transmission shifts. I didn't think it could be smoother than a top loader/Hurst, but it is.
Gear ratios are a little low, but the fix it to go to a 3.31 rear-end. This is with a 428, soon to be a 482. Every change gets closer to nirvana.

We just swapped my TKO600 to a T56. I currently have a 3.27 rear end but plan to do a gear swap. We intend to drive the car to see where the new cam wants to be before committing to a specific gear. I've got a set of 3.90 and 4.30..... we are also gong from a 315/35/17 to a 325/50/15. According to the gearing calculator the 4:30s put my setup similar to if I swapped to a 4:10 with my current tire size. Will be running a 575hp 427 Windsor with drag radials on the rear setup for the street.

Also have a Richmond street 5 speed part # 7041710 was installed in car by shell valley as a assembled kit when purchase by previous owner in 2002 and has yet to have ever been on road so no miles as of yet.

TKO 600 wr od
3.08 gears, so I'm at 1500 rpm at 60 mph with the .64 od. I have the 17" wheels which are a little smaller than the 15s.

I have a toploader behind my 289... Came with the car.
I thought I would be swapping it out for a WC T-5, but nope... It's not gonna happen.
The toploader reminds me of the Muncie M22 I used to have in my 69 Corvette (both shift like butter, but the toploader is WAY quieter )
with 3.31 gears out back, I can 0-60 in first gear (4.79 is my best so far, according to my Speedhut GPS speedo ), and I can still cruise in 4th at 75mph, turning 2800rpm, and getting 20+ mpg... It's true love.

I have a tko600, 3:55 ass end FE 445. About 2200 rpm in 5th at 60 I think. Its like an automatic... I can go down to about 1200 rpm coming up to a stop light and just roll on
